Summary

The evidence for pairing caffeine with L-theanine is stronger for attention-switching than for alertness or reducing jitteriness. A 2025 systematic review of roughly 50 trials of tea, L-theanine, and combined L-theanine plus caffeine found a small-to-moderate, significant combination benefit for attention-switching accuracy; point estimates also favored the combination for alertness, but those findings were not significant. The most direct test of the jitteriness claim, a 48-person trial, found L-theanine blunted caffeine's rise in blood pressure but did not significantly reduce self-rated jitteriness. Does caffeine plus L-theanine actually improve focus and cut the jitters?

The honest answer splits in two, and even the stronger half is narrower than the marketing suggests. A 2025 review covering roughly 50 trials of tea, L-theanine, and caffeine-plus-L-theanine found a significant combination-versus-placebo benefit for attention-switching accuracy; the parallel alertness claim pointed the same direction but didn't reach significance.1 For the more marketed claim — that L-theanine "cancels out" caffeine's jitteriness — the best direct test is more mixed: a 48-person trial found L-theanine blunted caffeine's blood-pressure rise but didn't significantly reduce jitteriness itself.2

I formulate MIHIYO's Energy-Focus oral dissolving strip (ODS) — a thin polymer film that dissolves against the oral mucosa without water — around caffeine and L-theanine, so I read this literature separately for each claim rather than treating "the stack works" as one fact. They are not the same claim, and not equally well supported.

Why the two ingredients are paired in the first place

Caffeine and L-theanine act through different mechanisms that may converge on some aspects of attention. Caffeine is a competitive antagonist at adenosine A1 and A2A receptors — adenosine signals the brain to wind down, so blocking its receptors is what makes caffeine feel alerting. That mechanism, at higher doses, also produces jitteriness: in the Rogers trial, a single 250 mg dose increased both self-rated jitteriness and blood pressure versus placebo.2

L-theanine is a non-protein amino acid found naturally in tea leaves. It does not block adenosine; its precise human mechanism is uncertain, though animal/in vitro research links it to GABA/glutamate signaling.3 In a small human EEG study, a single 50 mg dose was linked to increased resting alpha-band activity, a pattern associated with calm, wakeful alertness.4 The hypothesis: caffeine supplies drive through one receptor system, L-theanine a calming shift through another, and the second in theory smooths the first's edges without erasing alertness — though this rests on firmer ground for caffeine's well-characterized human pharmacology than for L-theanine's, where the GABA/glutamate evidence is still mostly animal and in vitro.

That hypothesis holds up better for attention-switching than for alertness or jitteriness, as the next two sections show.

What controlled trials show for attention

This is the better-supported half of the literature. The most comprehensive recent review is a 2025 Nutrition Reviews systematic review of approximately 50 randomized trials of tea, L-theanine alone, and L-theanine-plus-caffeine, with 15 to 16 contributing to at least one meta-analysis. For the combination, attention-switching accuracy improved significantly versus placebo in both the first hour (P=.004) and second hour (P=.008) after dosing, and digit-vigilance accuracy improved significantly in the second hour (P=.04). Alertness point estimates favored the combination too, but both hours' confidence intervals crossed zero, so neither was statistically significant.1 Most pooled analyses compared the combination with placebo, not caffeine alone. An earlier meta-analysis attributed much of the benefit to caffeine, and this review's own comparisons of L-theanine alone versus caffeine alone favored caffeine too, though imprecise and non-significant — so L-theanine's incremental contribution remains unresolved.1

A newer 2026 meta-analysis pooled 31 trials (1,168 participants, healthy and clinical) of L-theanine against placebo, not the combination; a single 200 mg dose significantly improved choice reaction time in healthy adults (SMD 0.51), with a smaller stress effect driven largely by high-risk-of-bias studies.5 That supports an acute reaction-time effect for L-theanine, not that it improves caffeine's own effect.

Individual trials fill in the picture. A crossover trial using 50 mg caffeine with 100 mg L-theanine found the combination improved attention-switching accuracy and reduced distractibility more than placebo in 27 adults.6 A separate trial using 40 mg caffeine with 97 mg L-theanine found the combination improved task-switching accuracy and alertness while reducing tiredness in 44 young adults — and produced a small but significant systolic blood-pressure increase versus placebo, with a nonsignificant diastolic trend; heart-rate findings were unclear.7 A 2025 trial in 37 sleep-deprived adults found 200 mg L-theanine plus 160 mg caffeine improved selective-attention accuracy, discrimination, reaction time, and P3b measures versus placebo; with no caffeine-only arm, it shows the combination beats placebo, not that L-theanine adds to caffeine.8

A small 2020 proof-of-concept trial tested the combination in a four-way crossover of five male completers with ADHD, ages 9.5–15.2 (one of six recruited withdrew before dosing), and found significantly improved sustained attention (Go/NoGo d-prime, p=0.033) versus placebo. But on that same task, the default-mode-network reduction — the mind-wandering circuit — was significant for L-theanine alone and caffeine alone, not the combination (though significant for the combination on a separate stop-signal task). The result is real; a five-completer trial still needs replication.9

Several trials include a caffeine-only comparator, with mixed results. Haskell's 2008 trial (150 mg caffeine, 250 mg L-theanine) found a significant caffeine-by-L-theanine interaction on word-recognition speed, plus faster reaction time, working-memory speed, and sentence-verification accuracy beyond either alone.12 Dodd later found adding 50 mg L-theanine to 75 mg caffeine eliminated some of caffeine's benefits at that lower ratio, while the small 2026 Arslan trial found improvements in auditory reaction time and gaming precision when L-theanine was added.1011 The literature hasn't established an optimal ratio — among trials in the 2025 meta-analyses, median doses were 82 mg L-theanine and 88.74 mg caffeine, and no trial has isolated ratio as the cause of a positive or negative result.1

Where the jitteriness claim gets thinner

This is the part of the pairing repeated as settled fact more often than the data supports. "L-theanine cancels out caffeine's jitters" sounds specific and tested; the best direct evidence says it's only half right.

The clearest evidence comes from a 2008 trial that gave 48 healthy adults 250 mg caffeine, 200 mg L-theanine, the combination, or placebo, and rated mood — including jitteriness — alongside blood pressure. Caffeine alone raised both jitteriness and blood pressure. L-theanine alone changed neither significantly versus placebo. In combination, L-theanine blunted caffeine's blood-pressure rise — but didn't significantly reduce the jitteriness.2 That doesn't fully support the popular claim.

Giesbrecht's 2010 trial found that at a different dose — 40 mg caffeine plus 97 mg L-theanine — the combination produced a small but significant systolic BP increase versus placebo, with a nonsignificant diastolic trend. Without a caffeine-only arm, it can't show whether L-theanine attenuated, amplified, or had no effect on caffeine's own response — only that the combination differed from placebo.7 That sits alongside Rogers' finding that L-theanine blunted caffeine's BP rise at a different dose — this literature hasn't resolved how dose shapes the picture. A related but frequently misread study tested L-theanine and caffeine in separate arms: in participants whose BP rose most under placebo, L-theanine significantly attenuated that rise, and across the full sample significantly reduced Tension-Anxiety. Caffeine showed only a nonsignificant BP trend and no Tension-Anxiety reduction.13 That's limited support for L-theanine's calming pharmacology alone — the sample was small and the BP finding came from a subgroup.

One trial pitted the combination against each ingredient alone under emotionally arousing conditions and found something more complicated than "cancels out the jitters." Caffeine and L-theanine pushed visual attention in opposite directions alone — caffeine broadened it, L-theanine narrowed it — and combined, the effects offset so thoroughly the combination didn't differ from placebo.14 Offsetting isn't the same as improving.

A January 2026 crossover trial in 20 athletes tested caffeine, L-theanine, and the combination on strength, endurance, and coordination; it found no additive benefit, and its authors said the results "challenge the prevailing assumption" of synergy.15 It didn't measure jitteriness, so it argues against broad synergy, not calming effects. It's also no longer the newest: an April 2026 esports trial found a real edge over caffeine alone (above),11 and a 2026 trial in 21 ADHD adolescents found both the combination and methylphenidate reduced false alarms and improved P3b versus placebo, though only methylphenidate improved overall reaction time — with no caffeine-only arm, and a pediatric sample that shouldn't generalize to healthy adults.16 The newest evidence is mixed and task-dependent.

What this means for MIHIYO's Energy-Focus strip

I hold every MIHIYO formulation to two metrics: bioavailability and time to peak plasma concentration — though this ODS has no published human PK data, so those are design considerations, not measured properties. That's why the Energy-Focus ODS pairs caffeine with L-theanine at or above the caffeine dose, following Owen's and Giesbrecht's positive trials — not a claim that an optimal ratio is established.

The formulation trade-off is real: two actives in one film leave less room for either dose before it gets too thick to dissolve within its target 30-to-60-second window. I size caffeine to the moderate range used in trials with a positive signal, leaving room for L-theanine, not a symbolic amount.

What I do not put on the label is a promise the strip eliminates jitteriness — the most direct trial found no significant reduction versus caffeine alone,2 and a decision built on evidence that holds up is one I can stand behind. Where this pairing falls short

Several honest limits apply. First, an optimal dose ratio has not been established, and trials with caffeine-only comparators have produced mixed, task-dependent results.101112 Second, the benefit is context-dependent: under emotional arousal, the two ingredients' opposing mechanisms can offset entirely rather than combine constructively.14 Third, the jitteriness evidence cuts against the popular claim: the best trial found L-theanine blunted caffeine's BP rise but not its jitteriness; a second found the combination's BP rose versus placebo, though without a caffeine-only arm to isolate L-theanine's effect.27 Fourth, most attention evidence compares the combination with placebo, not caffeine alone — L-theanine appears to have attention effects on its own, but not consistently added to caffeine's.15 Fifth, sample sizes are frequently small — five completers in the ADHD trial, 15 in the esports trial, 20 in the athlete trial — and none used an oral dissolving strip. Sixth, industry involvement is common here: the 2025 review discloses Lipton- and Unilever-affiliated authors and industry as its most common funding source, and Giesbrecht's trial was run entirely by Unilever R&D — not disqualifying, but it strengthens the case for independent replication.17

The bottom line

The combination beats placebo on some acute attention measures, particularly attention-switching accuracy, but whether L-theanine adds to caffeine's own benefit is uncertain and task-dependent — Dodd found it didn't, Arslan found it did — and the alertness claim wasn't significant in the 2025 analysis.11011 The idea that L-theanine specifically neutralizes caffeine's jitteriness is a plausible mechanism dressed as settled fact; the most direct trial found it blunted caffeine's blood-pressure rise but not the jitteriness itself, and a second found a small systolic increase versus placebo, without showing how that differed from caffeine alone.27 MIHIYO Labs builds its Energy-Focus strip around the attention-switching evidence, not an established ratio or a calming effect the evidence hasn't confirmed.
